ISOLATION AND IDENTIFICATION OF CHLOROPHYLLIDE BY HPLC-CHROMATOGRAPHY, VISIBLE AND MASS SPECTROSCOPY

In this work is represented the isolation method as well as an application of the Visible Spectrometry, High Pressure Liquid Chromatography and Electrospray Ionisation-Mass Spectrometry analysis for the identification of isolated and purified chlorophyllide. Chlorophyllide is a derivative of chlorophyll, namely, chlorophyll without phytil-chain in C-173 position. It is formed in the last steps of chlorophyll biosynthesis, or in degradation processes like senescence of leaves or in the reactions catalysed by several enzymes. In this work, at the in vitro conditions, the chlorophyllide is obtained using a pigment extraction from the dried plant material (Spinacia Oleracea L), coupled with the column chromatography. For identification of chlorohyllide are used tree analytical methods: Visible Spectrometry, High Pressure Liquid Chromatography and Electrospray Ionisation-Mass Spectrometry. The main components in the chlorophyllide fraction were identified as chlorophyllide a/b. Spectrophotometrically determined concentration ratios of the main identified pigments, chlorophyllide a/b (in the chlorophyllide fraction) were 6.89/1. These analytical methods are proved to be suitable for the identification of the chlorophyllide as well as other structurally similar pigments.
