One of the major efects that metals generates in biological systems is the production of free radicals which consequently leads to oxidative stress. Metal-induced oxidative stress then affects biomolecules such as lipids, proteins and DNA, thus contributing to numerous diseases including diabetes, atherosclerosis, cancer and neurological disorders. Redox active metals (Fe, Cu, Cr, Co) undergo redox cycling reactions and generates reactive oxigen speaces (ROS) via Fenton reaction, while redox inactive metals (Cd, As and Pb) indirectly induce oxidative stress through depletion of glutathione and bonding to sulfhydryl groups of proteins. Copper and zinc are antagonistically associated with Alzheimer’s disease: copper has neurodegenerative, and zinc neuroprotective role. In cadmium induced apoptosis imporant mechanism is oxidative DNA damage which is also related to chromium toxicity and intravenous iron administration. Current therapeutic approach involves antioxidants based on their ability to chelate metals and inhibit their participation in free radical reactions as well as to scavenge free radical intermediates.
